I have created a tiny jpeg called mini.jpg and I would like to use it as EXIF thumbnail of a file
named destination.jpeg, which has EXIF information, but without thumbnail.

I have tried
exiftool -ThumbnailImage<=mini.jpg destination.jpg
exiftool '-ThumbnailImage<=mini.jpg' destination.jpg
exiftool \"-ThumbnailImage<=mini.jpg\" destination.jpg

but none works.
What's the correct syntax?

Try this:

exiftool "-thumbnailimage<=mini.jpg" destination.jpg
